data-contrast=\"none\">Na d\u00e9cada de 1940, o cientista Maur\u00edcio Rocha e Silva, ent\u00e3o pesquisador da Faculdade de Medicina de Ribeir\u00e3o Preto (USP), identificou que o veneno da serpente <\/span><i><span data-contrast=\"none\">Bothrops jararaca<\/span><\/i><span data-contrast=\"none\"> induzia a libera\u00e7\u00e3o de uma subst\u00e2ncia a partir de prote\u00ednas plasm\u00e1ticas. Essa subst\u00e2ncia, at\u00e9 ent\u00e3o desconhecida, foi nomeada bradicinina e caracterizada como um pept\u00eddeo com potente efeito vasodilatador e espasmog\u00eanico. Contudo, embora apresentasse semelhan\u00e7as com outras aminas vasoativas, sua a\u00e7\u00e3o se distinguia pela resist\u00eancia \u00e0 atropina e aos anti-histam\u00ednicos, sugerindo um mecanismo fisiol\u00f3gico at\u00e9 ent\u00e3o in\u00e9dito.\u00a0<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335559685&quot;:-5,&quot;335559739&quot;:249,&quot;335559740&quot;:268,&quot;335559991&quot;:10}\">\u00a0<\/span><\/p><p><span data-contrast=\"none\">Estudos posteriores demonstraram que a bradicinina \u00e9 gerada pela a\u00e7\u00e3o de enzimas proteol\u00edticas sobre o cininog\u00eanio e que possui meia-vida curta, sendo rapidamente degradada por enzimas presentes no endot\u00e9lio \u2014 em especial a ECA, uma carboxipeptidase dependente de zinco.<\/span><b><span data-contrast=\"none\">\u00a0<\/span><\/b><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335559685&quot;:-5,&quot;335559739&quot;:249,&quot;335559740&quot;:268,&quot;335559991&quot;:10}\">\u00a0<\/span><\/p><p><span data-contrast=\"none\">Nas d\u00e9cadas seguintes, o farmacologista S\u00e9rgio Henrique Ferreira, tamb\u00e9m da Faculdade de Medicina de Ribeir\u00e3o Preto (USP), aprofundou essa linha de pesquisa e identificou, no veneno da <\/span><i><span data-contrast=\"none\">Bothrops jararaca<\/span><\/i><span data-contrast=\"none\">, um conjunto de oligopept\u00eddeos com a\u00e7\u00e3o sin\u00e9rgica \u00e0 da bradicinina: os fatores de potencia\u00e7\u00e3o da bradicinina (BPFs). Esses pept\u00eddeos mostraram-se capazes de inibir a ECA, ao mesmo tempo em que preservavam a bradicinina circulante e impediam a convers\u00e3o de angiotensina I em angiotensina II \u2014 um potente vasoconstritor end\u00f3geno.\u00a0<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335559685&quot;:-5,&quot;335559739&quot;:249,&quot;335559740&quot;:268,&quot;335559991&quot;:10}\">\u00a0<\/span><\/p><p><span data-contrast=\"none\">Essa a\u00e7\u00e3o dupla, comprovada em diversos modelos experimentais, resultava em queda significativa da press\u00e3o arterial, mesmo na presen\u00e7a de est\u00edmulos hipertensores. Um dos pept\u00eddeos mais potentes, o BPP<\/span><span data-contrast=\"none\">\u2089<\/span><span data-contrast=\"none\">a, demonstrou alta afinidade pela ECA e expressivo efeito hipotensor. Com base nessas estruturas, S\u00e9rgio Ferreira, em colabora\u00e7\u00e3o com John Vane, sintetizou o pentapept\u00eddeo PCA-Lys-Trp-Ala-Pro (BPP<\/span><span data-contrast=\"none\">\u2085<\/span><span data-contrast=\"none\">a), que viria a inspirar o desenvolvimento do captopril.\u00a0<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335559685&quot;:-5,&quot;335559739&quot;:249,&quot;335559740&quot;:268,&quot;335559991&quot;:10}\">\u00a0<\/span><\/p><p><span data-contrast=\"none\">O composto final, desenvolvido pela empresa Squibb (hoje Bristol-Myers Squibb), manteve a extremidade terminal de prolina e incorporou um grupo tiol \u2014 elemento essencial para sua alta afinidade ao centro catal\u00edtico da ECA. O captopril tornou-se, assim, o primeiro inibidor competitivo da ECA com efic\u00e1cia cl\u00ednica comprovada.<\/span><b><span data-contrast=\"none\">\u00a0<\/span><\/b><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335559685&quot;:-5,&quot;335559739&quot;:249,&quot;335559740&quot;:268,&quot;335559991&quot;:10}\">\u00a0<\/span><\/p><p><span data-contrast=\"none\">Al\u00e9m de reduzir a convers\u00e3o de angiotensina I em II, o captopril aumenta a biodisponibilidade de pept\u00eddeos vasorreguladores como a angiotensina-(1\u20137), que atua no eixo contrarregulador do sistema renina-angiotensina. Embora o f\u00e1rmaco tenha sido sintetizado por uma farmac\u00eautica internacional, os fundamentos bioqu\u00edmicos e fisiol\u00f3gicos que possibilitaram sua cria\u00e7\u00e3o \u2014 da caracteriza\u00e7\u00e3o da bradicinina \u00e0 elucida\u00e7\u00e3o da a\u00e7\u00e3o dos BPFs \u2014 foram estabelecidos no Brasil, na Faculdade de Medicina de Ribeir\u00e3o Preto (USP).\u00a0<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335559685&quot;:-5,&quot;335559739&quot;:249,&quot;335559740&quot;:268,&quot;335559991&quot;:10}\">\u00a0<\/span><\/p><p><span data-contrast=\"none\">Essa trajet\u00f3ria exemplifica como abordagens interdisciplinares e o estudo de mol\u00e9culas bioativas oriundas de fontes naturais podem gerar solu\u00e7\u00f5es com alto valor agregado.
