O DNA reage com diversos metab\u00f3litos celulares, como alde\u00eddos e esp\u00e9cies reativas de oxig\u00eanio, diferentes agentes qu\u00edmicos e f\u00edsicos ao nosso redor, como componentes do cigarro, radia\u00e7\u00e3o ultravioleta do sol ou radioatividade, al\u00e9m da pr\u00f3pria \u00e1gua, de forma que o DNA precisa ser constantemente reparado da maneira mais fiel poss\u00edvel para evitar o ac\u00famulo de muta\u00e7\u00f5es que levam ao c\u00e2ncer, envelhecimento e neurodegenera\u00e7\u00e3o.<\/p>\n<p style=\"color: #000000; text-align: justify;\"><\/p>\n<p style=\"color: #000000; text-align: justify;\">N\u00f3s estudamos as vias moleculares de sinaliza\u00e7\u00e3o e reparo destes danos ao DNA, utilizando t\u00e9cnicas de biologia molecular e celular, por exemplo Western Blotting, microscopia de fluoresc\u00eancia, tecnologia CRISPR\/Cas9 e ensaios funcionais. Dentro dessa linha de investiga\u00e7\u00e3o, os projetos em desenvolvimento no laborat\u00f3rio se encaixam basicamente em tr\u00eas \u00e1reas de interesse:<\/p>\n<p style=\"color: #000000; text-align: justify;\"><\/p>\n<p style=\"color: #000000;\"><!-- \/wp:paragraph --><!-- wp:paragraph {\"customTextColor\":\"#000000\",\"fontSize\":\"normal\"} --><\/p>\n<p style=\"color: #000000; text-align: justify;\"><strong>ADP-ribosila\u00e7\u00e3o e reparo de DNA <\/strong><br>ADP-ribosila\u00e7\u00e3o \u00e9 uma modifica\u00e7\u00e3o p\u00f3s-traducional de prote\u00ednas com importante fun\u00e7\u00e3o na sinaliza\u00e7\u00e3o da presen\u00e7a de danos ao DNA, remodelamento da cromatina e recrutamento de fatores de reparo de DNA para o local da les\u00e3o, al\u00e9m de um tipo de morte celular programada chamada <em>parthanatos<\/em>. Esta modifica\u00e7\u00e3o \u00e9 catalisada por uma classe de enzimas chamadas poli(ADP-ribose) polimerases (PARPs), diversas das quais s\u00e3o ativadas por danos ao DNA. Centenas de prote\u00ednas s\u00e3o modificadas por ADP-ribose, outras tantas reconhecem e se ligam a esta modifica\u00e7\u00e3o, e outras mais s\u00e3o envolvidas na remo\u00e7\u00e3o deste \u201csinal\u201d ap\u00f3s a conclus\u00e3o do reparo. N\u00f3s buscamos entender os mecanismos moleculares pelos quais cada um desses eventos sinaliza a presen\u00e7a de danos ao DNA e acelera o seu reparo. Estas doen\u00e7as s\u00e3o geralmente caracterizadas por predisposi\u00e7\u00e3o ao c\u00e2ncer, envelhecimento precoce, imunodefici\u00eancia e\/ou problemas neurol\u00f3gicos, como neurodegenera\u00e7\u00e3o ou microcefalia. Alguns v\u00edrus, incluindo o coronav\u00edrus, expressam uma enzima cuja fun\u00e7\u00e3o \u00e9 hidrolisar (ou seja remover) a ADP-ribosila\u00e7\u00e3o de prote\u00ednas, que \u00e9 catalisada por enzimas do hospedeiro como parte da resposta antiviral das c\u00e9lulas.
